Improved morphological preservation with the protective recovery method

        A direct comparison of the traditional sucrose protective cutting method and the NMDG protective recovery method was performed using 5 month old mature adult mice.  Acute brain slices were prepared and IR-DIC images acquired at ~5 min and ~30 min following the slicing procedure to evaluate the extent of morphological preservation.  This approach revealed a dramatic improvement with the NMDG protective recovery method, with much less initial neuronal swelling evident at 5 minutes and much reduced neuronal shrinkage at 30 minutes post slicing.  Thus, the number of neurons that appeared accessible for patch clamp recordings was greatly increased.​
